Z-Leu-Leu-Glu-AMC is a fluorogenic tripeptide substrate used to measure postacidic-like (peptidylglutamyl-peptide-hydrolyzing) activity of the 20S proteasome. Proteolytic cleavage releases 7-amino-4-methylcoumarin (AMC), producing a fluorescent signal suitable for proteasome activity assays, kinetic studies, and inhibitor screening.
